Proliferative vitreoretinopathy, is a potentially blinding condition and is the main complication following retinal detachment. Complicated cases of retinal detachment are treated with silicone oil (SiO) tamponades which can potentially be used as drug reservoirs. This provides an opportunity for SiO tamponades to have an adjunctive, drug-releasing action at clinically relevant timescales.
This talk will describe the development of two SiO additives; chain-end modified PDMS and a series of linear amphiphilic copolymer architectures.  The effects of modified PDMS additives in SiO were investigated and have shown a positive effect on both drug solubility and longevity of release.  Different compositions of linear amphiphilic copolymers were also successfully synthesised and displayed high solubility within SiO. Studies of drug release using traditional UV-Vis and radiochemical approaches show that the presence of a linear graft copolymer can extend release as well as alter the release profile of drugs from the silicone oil phase. Finally cytotoxicity of these materials has been evaluated using an adult retinal pigment epithelium cell line and analysis of drug release has been demonstrated using a newly developed ex-vivo model.
Helen is currently a postdoctoral research associate in both the Departments of Chemistry and Eye & Vision Science at the University of Liverpool. Her research encompasses radiolabelled polymers, drug delivery for both the anterior and posterior segments of the eye and development of drug delivery models; in vitro and ex vivo. Before taking on this role Helen completed her PhD at the University in 2015. Her PhD work focussed on polymeric materials for ophthalmic drug delivery, where she developed an understanding of how the requirements and restrictions from
both disciplines are vital to progress. Her research interests include optimising biocompatibility of polymeric materials and enhancing their capability of drug delivery.
